Obviously not counting games with character creation (and even in those the max age slider makes the character look like a 30-year-old with bad skin).

Lara Croft was 30 in the first tomb raider and by the end of the first generation of games was near 40 iirc?
If you've played the cool little tactics/stealth game Invisible Inc the main character is an older woman who runs the spy agency. You have to unlock her to add her to your team of playable characters I believe, but she's definitely the main focus.
